I'm doing some ILs in the DLC category (which must include builds from the DLC, i.e. Chalice, weapons or charms from the DLC). However, I've seen a lot of WR of these categories with no builds from the DLC, for example, runs with just mugman, charge-shot and wetstone. This is seen more cleary in plane levels, uploaded in the DLC category, but the build is Cuphead and Wetstone. Am I missing something?

Of course you can use cuphead/mugman, but you need to use at least one DLC weapon or charm

Both of the runs you linked are using the Divine Relic charm, some people also use Heart Ring for plane levels or Twist-up as secondary weapons in ground levels
