I don't have a version with offers DLC, so I sadly can't test the following question by myself and every YT-video i found is only showcasing the bonus characters after completing the game (buying everything etc.) with 100%.

So, in point of available spells: Are the extra characters like Godric Gryffindor handled as students (all at the same leaning level as Harry & Co., no matter how old/in which grade they are) or as teachers (which always have every spell, but only can be collected -token- and bought -shop- and used -freeplay/hogwarts- after learning reducto/completing the story)? Because if yes, if they are grown-ups like teachers, then you would have every spell available since the beginning, after you get just a little bit of money to purchase one of them. I don't know, if it actually would revolutionize the speedrunning aspect of the game, as you still have to do everything (somewhen: 100%, or as a fast rush through: any%), but at least for casual gaming, that'd somehow break the inner game's logic, right?

Hey, the DLC comes with the LEGO Harry Potter Collection version of the game and isn't accessible on the PC version. It is allowed to use the founders characters in 100% runs on LHPC so yes, the route difference from the Original version would differ drastically.
