To answer your question, each speed run starts from when the first frame of the game screen appears, in other words after you press the "play" button.

I again made a private youtube video so that I could time this correctly.

Please look into exporting the origional video from screencastify so that you can upload it to you tube and submit the videos that way, thank you.
