The basis of Any% is to end the game as quickly as possible with a basic illness. If a person finishes the game faster with a disease other than Neurax Worm, they fulfill the conditions of Any%. There are no usage restrictions other than the basic rules.
Your remark is interesting but generally we don't speedrun a game without having tested it and without having trained before. The runners will therefore logically unlock all the content of the game. So the "NG +" is a standard for a good run.
The game has been on speedrun.com for 5 years. Since I am a moderator (4 years old), the rules have greatly evolved and many categories have been added. It may be possible that one day an update of the rules will be made, but for now they are very good like that. The game is very accessible compare to many others.

If the basic rules are not followed, your run will not be accepted. For the rest, it's up to you to find a way to beat your score or that of others !

Yes I had thought of doing that. But the campaigns where to go beat the chaos is very difficult. Because it appears randomly and takes a long time to come. The speedrun still has the goal of finishing a goal as quickly as possible. So I find it normal to play the game x3. But for each quests I add: "- Play at the speed of your choice." That way, the person who thinks he can do better in x1 or x2 can do it
