After the first 24 hours of testing, that's what we collected on the Discord channel (between all the runners)
- First 3 areas are pretty much straightforward, I'd say Cassandra's (Hollow) then Vito's (River Row) then Burke's (Pointe Verdun) ... then personally I went Thickfaw Harbor and Southdowns for immediate proximity, maybe other runners took different strats, don't think changes too much since most of the missions are inside the relative area

Death Warp: Dying after completing missions will warp you to your area boss (depends on the area), consider this while routing, could save driving time, especially we can test (and hopefully exploit) this when we need to leave Bayou, that's way far away from anything else

Door unlocking: after the first part of the minigame (finding the green short area for unlocking), spamming E seems to solve the second part of the minigame pretty much most of the times ... if it doesn't work, you lose like 1 second and you can retry normal way.

Window clipping glitch (could be worth in a couple of missions, will depend on route/strategy)

Death feel random sometimes, in Bayou area i got spawned not in the city but somehere else where i was before. I think it spawns where your game saved last or smth.
Here's a fast and safe way for the Tanager part in Harbor that wall-climb at the end might be useful in other places as well, I think it works because the corner bit doesn't have spikes.
also an example way to kill frank before chase starts, this one might have a million ways to do
--Edited for improved route, faster and safer-- There's a way to skip all gunfights in casino there's still some fine tuning on that, which wall sections are easiest to get through etc. but the basic route is pretty much there. The guards seem to differ their position in their routes a little, might be just from checkpoint-loading while testing. In any case, there's 3 to kill, 1 down and 2 up. You have to do the first clip and visit hallway for a trigger.
You can copy your save files for easier testing/practise. Go to %LOCALAPPDATA%\2K Games\Mafia III\Data"longnumber"\gamesaves\default\aslot. there's 3 files in there, named save0-2.sav. just copy those somewhere else. when you want to load a save, just exit the game and copy your backups to "aslot" folder and replace whatever's in there. HOWEVER, these files don't include collectibles, if you overwrite your saves the collectibles will reset.
Here's a copypasta from few notes from my 5:47 run, so they don't get lost in discord history.
the basic route is the same, but I did a couple load-warps and more efficient pickups in rackets the lack of tutorial pop-ups save a lot of time, especially in earlier splits. and my old one had all sorts failing, from both me and the game. Reload after every time you give a racket/district to someone. (the last one might have a risk of infinite load, needs testing). Generally the checkpoint load puts you in the nearest "safehouse", which usually means closest mission giver or taken over rackethouse. But it won't warp you to npc that you haven't met yet. Warping might also lose you some racket dmg, since the game doesn't save after every pickup. It does save when completing an objective (see hollows below), and probably when you complete the dmg required for racket (haven't tested but it happened with southdowns gambling on my run).
In hollows prostitute racket, leave one of the doors (where you save the 3 whores) until you have 7k or less damage left in the racket. Then kick the last door, it saves the game and leaves you with 5k left to do. Then you reload checkpoint, which puts you outside Donovan. Talk to him and then reload again, which warps you to the Voice. Take a shortcut through a window in the riverboat, just shoot the window out and jump through. It only works with that window because there's a couch in front of it.
and here's the overall order I used, everything fits together so I never really tried other routes.
